LUCIDITY Trial Results and Safety Profile
Amylyx said in a press release on Aug. 18, 2026, that LUCIDITY was a 78-participant, multicenter, randomized, double-blind, placebo-controlled Phase 3 trial in adults with post-bariatric hypoglycemia following Roux-en-Y gastric bypass surgery. The investigational drug, avexitide, is a first-in-class glucagon-like peptide-1 (GLP-1) receptor antagonist.
The company reported that LUCIDITY met the FDA-agreed primary endpoint, showing a 55% reduction in the composite rate of Level 2 and Level 3 hypoglycemic events compared with placebo (p=0.000003). All secondary endpoints were also met, including reductions in Level 2 hypoglycemic events measured by self-monitoring of blood glucose and continuous glucose monitoring, as well as Level 3 severe hypoglycemic events. These reductions were consistent, highly statistically significant, and clinically meaningful.
Avexitide was described as generally well-tolerated with a favorable safety profile.
Regulatory Path and Company Plans
Amylyx said it will submit a New Drug Application (NDA) to the FDA by year-end 2026 for avexitide in post-bariatric hypoglycemia following Roux-en-Y gastric bypass, based on the LUCIDITY efficacy and safety data. The drug holds FDA Breakthrough Therapy and Orphan Drug designations for this indication.
On its Aug. 6, 2026 second-quarter earnings call, management said LUCIDITY was powered at 90% to detect a 35% treatment effect and that topline data were expected in late August or early September.
